Objectives

The course is organized in face-to-face lessons integrated by laboratory activity that give the knowledge on eukaryotic cell structure and on
mechanisms, vesicle trafficking included, aimed at the maintaining of chemical difference of compartements and organelles, related to their functions. By analyzing cell processes related to differentiation, students learn the peculiarity of different tissues, examining in depth regulatory mechanisms of the cell cycle and iimplications of apoptotic cell death and
stem cell component in tissue turn-over.

Contents

MOLECULES and IONS in the cells. PLASMAMEMBRANE: lipids, proteins and glucides; TRANSPORT MECHANISMS. GLYCOCALIX: cell recognition and adhesion. RECEPTORS and SIGNAL TRANSDUCTION. NERVE IMPULSE. JUNCTIONS between epithelial/non epithelial cells and EXTRACELLULAR MATRIX of CONNECTIVE TISSUES. Endomembranes. ENDOPLASMIC RETICULUM. Ribosomes. GOLGI APPARATUS. LYSOSOMES, peoxisomes. Endocytosis and exocytosis; secretion types. MITOCHONDRIA.
CYTOSKELETON: microtubules, intermediate filaments, microfilaments and plasmamembrane interactions. CONTRACTILITY (muscle/non muscle cells). LEUCOCYTE MIGRATION and defence mechanisms. NUCLEUS: envelope, lamina; pores and trafficking; hetero-euchromatin, nucleolus. CELL CYCLE: phases and its regulation. APOPTOSIS.

Teaching Methods

Lectures and laboratory activity in which adherent cell cultures are submitted to fluorochromization procedures with vital probes, that are selectively uptaked by structures and organelles on the basis of a specific chemical affinity. These techniques allow students to analyse plasmamembrane structure and to evaluate functionality of cell organelles, such as nucleus, mitichondria and lysosomes. The use of specific DNA fluorochromes permit the analysis of proliferative activity and apoptotic events.
